Daraxonrasib trial shows doubled survival, EMA review ongo‑g
Entities: KRAS gene · mKRAS-VAX · daraxonrasib · pancreatic ductal adenocarcinoma
Overview
The rolling EMA review of the oral KRAS‑targeting inhibitor daraxonrasib, launched on 7 July 2026, continues as new clinical data emerge. A multinational phase‑III trial reported on 25 July that daraxonrasib doubled median overall survival in patients with metastatic pancreatic ductal adenocarcinoma, extending it to 13.2 months versus 6.7 months with standard chemotherapy. Progression‑free survival also improved (7.2 months vs 3.6 months) and the death rate was lower in the 248‑patient daraxonrasib arm compared with 252 patients receiving physician‑chosen chemotherapy. The drug, developed by Revolution Medicines, remains under regulatory evaluation and is not yet approved for general use. A follow‑up report on 31 July reiterated the survival benefit and described the drug’s mechanism as binding KRAS in its active (RAS‑ON) state to block downstream signaling. The same update highlighted an early‑stage trial of the KRAS‑mutant vaccine mKRAS‑VAX, which showed safety and robust T‑cell responses, with pancreatic cysts shrinking in some high‑risk volunteers. These vaccine findings are part of the broader KRAS‑targeting research landscape in the United States and Japan but have not altered the regulatory trajectory of daraxonrasib. The combined clinical and regulatory developments keep daraxonrasib at the forefront of efforts to improve outcomes for metastatic pancreatic cancer patients.
