Pancreatic Cancer Breakthroughs: Mitochondrial Inflammation Target and Survival‑Doubling Drug

Scientists at The Wistar Institute and ChristianaCare identified a vulnerability in pancreatic cancer cells involving loss of the mitochondrial protein Mic60. The resulting membrane leak releases double‑stranded RNA, which activates the TLR3‑TRAF6 inflammatory pathway that cancer cells depend on for growth. Blocking TLR3 or TRAF6 killed the tumor cells in vitro and halted tumor development in mouse models, while sparing healthy cells.

In a separate development, Revolution Medicines reported phase‑3 trial results for daraxonrasib, a KRAS‑targeted inhibitor. The drug extended median overall survival to 13.2 months, compared with 6.7 months for standard chemotherapy, effectively doubling life expectancy for patients with advanced pancreatic cancer. Daraxonrasib also produced tumor shrinkage and pain relief in many participants, highlighting its potential as a precision‑medicine approach for a disease that carries a 5‑year survival rate of just 13 %.
