Disc Medicine bitopertin regulatory and clinical status

Overview

Disc Medicine Inc. has faced scrutiny regarding its experimental treatment for erythropoietic protoporphyria (EPP), bitopertin. Following a February decision by the U.S. Food and Drug Administration (FDA) to decline approval and request more clinical evidence, several securities law firms launched investigations. These firms are examining whether the company provided materially misleading information to investors regarding regulatory risks prior to the FDA’s Complete Response Letter.

However, the company later reported successful results from its Phase 3 APOLLO trial. The study, involving 183 patients aged 12 and older across the United States, Canada, Europe, and Australia, met both co-primary endpoints. Results showed a 41.4 per cent reduction in whole-blood metal-free protoporphyrin IX (PPIX) compared to a placebo, alongside an average increase of 12.2 hours of pain-free light exposure per month. These findings specifically address previous FDA concerns regarding the link between PPIX reduction and clinical improvements in light tolerance.

Following this success, Disc Medicine plans to resubmit its New Drug Application (NDA) to the FDA by the end of 2026, with a regulatory decision expected by mid-2027. If approved, the once-daily oral medication could challenge the market monopoly held by Clinuvel Pharmaceuticals’ Cinere, which requires subcutaneous injections every two months.
