Amivantamab trial in advanced head‑neck cancer
Overview
In late May 2026, a multinational Phase II study of Johnson & Johnson’s sub‑cutaneous antibody amivantamab was presented at the ASCO meeting in Chicago. The trial enrolled 102 patients with advanced head‑and‑neck cancers across 11 countries. Results showed tumor shrinkage in 43 participants and complete disappearance in 15, yielding an overall response rate of about one‑third. Median overall survival was reported at 12.5 months and progression‑free interval around 6‑6.5 months, with most adverse events mild to moderate and fewer than 10 % discontinuing therapy.
Subsequent reports over the next days reiterated these findings, added details on median survival, highlighted comments from Professor Kevin Harrington, and quoted a patient experience. The data prompted the launch of larger Phase III studies (OrigAMI‑4, OrigAMI‑5) aiming to enroll up to 500 patients, with sites expanding to the United States, United Kingdom, Vietnam, Greece, Portugal and other regions. The drug, already approved for non‑small‑cell lung cancer, is also being investigated in around 60 trials for additional solid tumours such as lung, colorectal, brain and stomach cancers.
