Oligodendrocyte dysfunction and cognitive decline research

Overview

Research has identified a link between dysfunctional oligodendrocytes—cells responsible for producing myelin—and age-related cognitive decline. Studies involving the Lothian Birth Cohort 1936 indicate that individuals experiencing more severe cognitive decline show changes in nerve fibers and the presence of excess, unhealthy myelin.

Further investigation published in Nature Medicine utilized mouse experiments to explore these mechanisms. Researchers found that deleting the gene encoding nuclear factor erythroid 2-related factor 2 (NRF2) from oligodendrocytes in mice resulted in axon size alterations and myelin thickness changes similar to those observed in humans. These findings suggest that reduced NRF2 expression and oligodendrocyte density are key factors in cognitive impairment.
