Advancements in metastatic cancer clinical trials
Overview
Recent phase 3 clinical trials have demonstrated significant survival benefits for various metastatic cancer treatments. In gastroesophageal adenocarcinoma, the HERIZON-GEA-01 trial showed that zanidatamab combined with chemotherapy reduced the risk of death by 24% compared to trastuzumab plus chemotherapy for HER2-positive patients. The Clarity-Gastric01 study found that sonesitatug vedotin (Sone-Ve) reduced the risk of death by 42% in patients with Claudin 18.2 positive advanced gastric, gastroesophageal junction, or esophageal adenocarcinoma. Additionally, research into biliary tract cancer indicates that ivonescimab combined with gemcitabine and cisplatin showed a 25% reduction in the risk of death, while the HARMONi-GI1 trial reported a median overall survival of 14.9 months using ivonescimab with chemotherapy.
In the field of small cell lung cancer, the DeLLphi-305 Phase III trial found that combining tarlatamab with durvalumab reduced the risk of death by 58%.
New data presented at the European Society for Medical Oncology (ESMO) congress regarding endometrial cancer confirms the efficacy of sacituzumab tirumotecan (sac-TMT). In a phase 3 international trial published in ‘The Lancet’, sac-TMT—which targets the TROP2 protein—showed a median overall survival of 19.0 months compared to 12.1 months for standard chemotherapy. This represents a 32% reduction in the relative risk of death and a 41% reduction in the risk of disease progression or death. However, serious side effects were noted, with grade 3 or higher treatment-related side effects occurring in 54.4% of the sac-TMT group.
In ovarian cancer research, a phase 1 trial at the Dana-Farber Cancer Institute tested a personalized vaccine called NeoVax. When combined with the immunotherapy drug nivolumab, the vaccine successfully triggered a strong immune response in participants.
