Discovery of skull-based immune structures

Researchers from Washington University in St. Louis have identified previously unknown immunological structures located within the bone marrow of the skull. Published in the journal Nature, the study describes these formations as an “immune security station” or lymphoid structures similar to lymph nodes, primarily situated in the posterior regions of the skull, such as the occipital and interparietal bones.

The discovery challenges the long-held scientific belief that the brain’s immune system is largely isolated from the rest of the body. The research demonstrates a communication pathway where molecules from the brain can reach the skull bone marrow, which contains specialized immune cells including B cells, follicular helper T cells, and antigen-presenting cells. Scientists identified small channels leading from brain tissue through the dura mater directly into the skull, facilitating this connection.

In studies involving mice with glioma, an aggressive brain cancer, researchers found that inhibiting these skull-based immune structures led to faster tumor progression and lower survival rates. Lead author Jonathan Kipnis noted the skull bone marrow acts as a “safety barrier” in early stages of brain cancer. Strengthening these centers through specific proteins could provide new therapeutic avenues.

Subsequent research led by Harvard Medical School suggests these structures act as localized immune hubs that serve as rapid first responders. This activity may be linked to chronic pain and various neurological conditions. Specifically, elevated levels of the protein translocator protein (TSPO) have been observed near the skull bone marrow in individuals with migraines and visual aura. Scientists believe this activation might represent a neuroimmune response shared across conditions including Alzheimer’s disease, stroke, depression, and autism.
